Lack of causal effects or genetic correlation between restless legs syndrome and Parkinson disease

Estiar MA, Senkevich K, Yu E, et al. - Utilizing bidirectional Mendelian randomization and linkage disequilibrium score regression, researchers sought to explore evidence for a causal relationship and/or shared genetic architecture between Parkinson disease (PD) and restless legs syndrome. No evidence of a connection between restless legs syndrome and PD was discovered. Reverse Mendelian randomization also failed to show a causal effect of PD on restless legs syndrome. Linkage disequilibrium score regression analysis revealed that there is no genetic link between restless legs syndrome and PD. Confounding or nongenetic determinants could explain some of the associations observed in epidemiological studies.
