Interleukin 1 receptor antagonist (IL1RN) gene variants predict radiographic severity of knee osteoarthritis and risk of incident disease

Attur M, Zhou H, Samuels J, et al. - In this study, over 1,000 people who met American College of Rheumatology standards for tibiofemoral OA were chosen from three independent, National Institute of Health-funded cohorts, in order to investigate the correlation of single nucleotide polymorphisms of the Interleukin 1 receptor antagonist (IL1RN) gene with radiographic austerity of symptomatic knee OA and the risk of incident OA. It was found that the carriage of the IL1RN TTG haplotype was correlated with more critical rOA, raised risk for incident OA, and heightened evidence of inflammation in RA. Therefore, these data imply that the IL1RN TTG risk haplotype, correlated with reduced IL-1Ra plasma levels, impairs endogenous "anti-inflammatory" mechanisms.
