Genetic variation within the HLA-DRA1 gene modulates susceptibility to type 1 diabetes in HLA-DR3 homozygotes

Aydemir O, et al. - Multiple gene variants, environmental factors, and immunoregulatory dysfunction interact in type 1 diabetes (T1D), so researchers identified new T1D susceptibility loci using molecular inversion probe sequencing technology. In order to minimize HLA heterogeneity, T1D patients (N=365) and controls (N=668) homozygous for the HLA-DR3 haplotype were included. According to findings, the HLA-DRA1 tri-SNP haplotype could modulate class II expression, which suggests that greater T1D risk may be due to regulated class II gene expression. These outcomes may help to clarify the role of HLA in susceptibility to T1D and improve risk assessment of diabetes, especially in homozygous individuals with high-risk HLA-DR3.
