Genetic variants associated with T-cell mediated cutaneous adverse drug reactions: A PRISMA-compliant systematic review – an EAACI position paper

Oussalah A, Yip V, Mayorga C, et al. - Given a link of drug hypersensitivity reactions (DHRs) with high global morbidity and mortality, and human leukocyte antigens have been identified as predisposing factors, leading to the evolution of pharmacogenetic screening examinations, such as HLA-B*57:01 in abacavir therapy, which has successfully decreased the incidence of abacavir hypersensitivity reactions, researchers investigated genetic links that have been reported in DHRs, by performing this PRISMA-compliant systematic review. This review included 105 studies with 5,554 cases and 123,548 controls. These studies described genetic links with carbamazepine (n = 31), other aromatic antiepileptic drugs (n = 24), abacavir (n = 11), nevirapine (n = 14), trimethoprim-sulfamethoxazole (n = 11), dapsone (n = 4), allopurinol (n = 10), and other drugs (n = 5). Findings revealed that the most frequently described genetic variants related to DHRs were found to be located in human leukocyte antigen genes and genes implicated in drug metabolism pathways.
