CDHR3 asthma-risk genotype affects susceptibility of airway epithelium to rhinovirus C infections

Basnet S, Bochkov YA, Brockman-Schneider RA, et al. - Based on the information that CDHR3 (cadherin-related family member 3) represents a transmembrane protein highly expressed in airway epithelia and the only recognized receptor for rhinovirus C (RV-C), and severe exacerbations of asthma and increased susceptibility to RV-C infections has been reported in young children in correlation with a CDHR3 SNP (rs6967330) with G to A base change, researchers determined the subcellular localization of CDHR3 and investigated if epithelial cell function and susceptibility to RV-C infections of the airway epithelia are influenced by CDHR3 asthma-risk genotype. They found higher overall protein expression and RV-C binding and replication in relation to rs6967330 A allele vs rs6967330 G allele. Earlier ciliogenesis and higher FOXJ1 expression were observed in relation to the rs6967330 A allele. Membrane integrity or ciliary beat function was not significantly influenced by the CDHR3 genotype. Overall, these data offer information about the subcellular localization and likely functions of CDHR3 in the airways and showed an association between CDHR3 asthma-risk genotype and increased RV-C binding and replication.
