Effect of age on the relative efficacy of clozapine in schizophrenia

Jones R, et al. - Via performing a systematic review and meta‐analysis, researchers investigated the superiority of clozapine over alternative antipsychotic medication in adults with schizophrenia and examined if additional benefit could be achieved with its earlier initiation. Among 276 retrieved studies, 34 studies were included in the meta‐analysis. Outcomes support clozapine as superior to other antipsychotics. This effect seemed not convincingly affected by the age, although there was one study suggesting this effect. Because of the age of many of the assessed studies, and changes in reporting practice over time, they support undertaking new clozapine RCTs, which include age of illness onset as well as age at trial time, in order to gain meta‐analysable data for future use.
