Association of uveal melanoma metastatic rate with stochastic mutation rate and type of mutation

Szalai E, et al. - In this pooled data analysis, researchers studied the stochastic properties of primary uveal melanoma—including the mutation rate as a function of tumor size and metastatic rate relative to the type of mutation. Using previously published large data sets, the investigators computed the mutation rate in different sized uveal melanomas. Using the spherical cap method, they estimated tumor volume. A higher mutation rate was noted for smaller tumors, which might be explained by a greater number of cell divisions occurring during the expansion phase of smaller uveal melanomas. The first two peaks seem to be related to BAP1-mutated tumors, and the late peak seemed related to SF3B1-mutated tumors regarding time to clinically detected metastases.
