Association of glucose-lowering medications with cardiovascular outcomes: An umbrella review and evidence map

The Lancet Diabetes & Endocrinology Mar 04, 2020

Zhu J, Yu X, Zheng Y, et al. - For this umbrella review, researchers analyzed the connection between glucose-lowering medications and a broad range of cardiovascular outcomes and evaluated the strength of evidence for these relationships. PubMed, Embase, and the Cochrane Library were searched to identify systematic reviews and meta-analyses of randomized controlled trials investigating the cardiovascular safety of glucose-lowering medications. They identified six risk and 38 protective connections exhibiting a high strength of evidence. Six associations raised the risk of cardiovascular disease, including glimepiride, rosiglitazone, and pioglitazone. For the correlations between diabetes drugs and cardiovascular outcomes, the authors found varying rates of evidence; some drugs raised the risk of cardiovascular disease, while others showed benefit.
