Association of antihypertensive drug target genes with psychiatric disorders: A Mendelian randomization study

JAMA Psychiatry Jun 06, 2021

Chauquet S, Zhu Z, O’Donovan MC, et al. - Researchers aimed at examining the potential effects of different antihypertensive drug classes on schizophrenia, bipolar disorder, and major depressive disorder utilizing genetic data. In this genetic association study, a 2-sample mendelian randomization analysis was used. As per findings, lower genetically estimated angiotensin-converting enzyme (ACE) messenger RNA and protein expression, as proxies for ACE inhibitor treatment, were noted to be linked with raised risk of schizophrenia. In view of these findings, they emphasize necessity for greater pharmacovigilance and further investigation through pharmacoepidemiology studies into the effect of ACE inhibitors, especially those that are centrally acting, on psychiatric symptoms in cases with schizophrenia, as well as with incidence of late-onset schizophrenia.
