Varenicline for the treatment of postural and gait dysfunction in Parkinson disease (PD)

Hall D, Kapur S, Vaughan C, et al. - In this investigator-initiated, double-blind, placebo-controlled study, experts aspired to explore whether varenicline is effective for balance in Parkinson disease (PD). Participants with a PD clinical diagnosis were randomized for eight weeks to receive varenicline or placebo. Following dose escalation, participants were given 1 mg of drug twice daily until the end of the study. Patients with severe tremor have been eliminated. For this analysis, 36 candidates were randomized (82% men, 100% Caucasian). The findings did not imply that varenicline had an effect on balance in PD patients. Varenicline also did not appear to have an effect on cognition. Perhaps a study would reveal a difference between groups if an objective measure of balance had been used in place of the BBS. The authors do not, however, suggest further research.
