Thyroid stimulating hormone and bone mineral density: Evidence from a two-sample Mendelian randomization study and a candidate gene association study

Van Vliet NA, et al. - This research was designed in order to inspect if low circulating thyroid stimulating hormone (TSH) levels within the normal range were causally associated with bone mineral density (BMD). Additionally, experts evaluated if common genetic variants in the TSH receptor (TSHR) gene and genetic variants influencing the expression of TSHR (eQTLs) were related to BMD. It was determined that 706 common genetic variants were mapped to the TSHR locus and expression loci for TSHR. None of these genetic variants correlated with BMD at the femoral neck or lumbar spine. No evidence was obtained for a causal effect of circulating TSH on BMD. Furthermore, no connection was found between genetic variation at the TSHR locus or expression thereof and BMD.
