Prognostic significance of CXCR4 expression in acute myeloid leukemia

Du W, Lu C, Zhu X, et al. - In view of the significant role of CXCR4, CXC chemokine receptor 4 (CD184), in leukemia proliferation, extramedullary migration, infiltration, adhesion, and resistance to chemotherapy drugs, researchers analyzed the CXCR4 expression in 122 acute myeloid leukemia (AML) patients by flow cytometry. They identified significantly higher expression of CXCR4 in AML-M4/M5 vs other subtypes according to both FAB subtype and WHO classification. The high CXCR4 expression group had significantly higher FLT3 internal tandem duplication mutant. A Better prognosis was evident in correlation with low CXCR4 expression on AML cells, and the median overall survival was 318 days vs 206 days for low CXCR4 expression patients vs patients with high CXCR4 expression, respectively. CXCR4 expression, age, and extramedullary infiltration were identified as independent prognostic factors in multivariate analysis.
