Prognostic role and clinicopathological features of SMAD4 gene mutation in colorectal cancer: A systematic review and meta-analysis

Fang T, Liang T, Wang Y, et al. - In this systematic review and meta-analysis, researchers sought to explore the correlation of SMAD4 mutations with overall survival (OS) and progression-free survival (PFS)/recurrence-free survival (RFS), as well as the association between SMAD4 mutations and clinicopathological features of early and advanced colorectal cancer (CRC). In total, 4,394 patients were involved. According to the findings of this meta-analysis, SMAD4 mutation is associated with OS, PFS/RFS, and clinicopathological parameters such as tumor site, disease stage, RAS status, lymph node metastasis, and mucinous differentiation. The results indicated that SMAD4 mutations can predict poor prognosis and aggressive clinicopathological characteristics of CRC. More large-sample cohort studies are required to confirm this finding. Because SMAD4 mutations are closely linked to RAS mutations, their connection warrants further investigation.
