Overexpression of Notch3 is associated with metastasis and poor prognosis in osteosarcoma patients

Tang XF, et al. - In this clinicopathological analysis of 70 cases of osteosarcoma, researchers focused on the impact of Notch3 expression on the prognosis (mainly survival) of osteosarcoma patients, as well as its role and mechanism in osteosarcoma cells. They used osteosarcoma cell lines MTH and U2OS and evaluated the cell cycle, cell viability, and wound healing capacity following knockdown of Notch3 by lentiviral transfection and siRNA. They carried out the Transwell assay and used RT-PCR and Western blot assay to determine the expression levels of hairy and enhancer of split-1 (Hes1) and matrix metalloproteinase 7 (MMP7). A significant influence of Notch3 expression on patient survival was indicated by comparing survival curves. Notch3 was shown to be an independent prognostic factor for osteosarcoma in multivariate analysis. In addition, via the downstream target gene Hes1 and effector MMP7, Notch3 might regulate invasion and metastasis of osteosarcoma.
