Oral fluconazole use in the first trimester and risk of congenital malformations: Population based cohort study

Zhu Y, Bateman BT, Gray KJ, et al. - Researchers conducted this population based cohort study to investigate the risk of congenital malformations correlated with exposure to oral fluconazole at commonly used doses in the first trimester of pregnancy for the treatment of vulvovaginal candidiasis. The research cohort of 1,969,954 pregnancies contained 37,650 (1.9%) oral fluconazole exposed pregnancies and 82,090 (4.2%) first trimester exposure to topical azoles. Oral fluconazole use was not associated with oral clefts or conotruncal malformations in the first trimester, but an association with musculoskeletal malformations was found, corresponding to a small adjusted risk difference of around 12 incidents per 10,000 overall exposed pregnancies.
