Morphology, p16, HPV, and outcomes in squamous cell carcinoma of the penis: A multi-institutional study

Eich ML, del Carmen Rodriguez Pena M, Schwartz L, et al. - A total of 102 persons were involved in the study in order to assess the pathologic characteristics and clinical outcomes in cases of invasive penile squamous cell carcinoma (SCC) and the correlation with p16 immunohistochemistry (IHC) and human papilloma virus (HPV) in situ hybridization (ISH). Tumor histology associated well with p16 positivity and in 25/26 cases, the presence of HPV was precisely prognosticated p16 IHC. On multivariate analysis, PNI was related to local disease recurrence whereas LVI was correlated with progression to metastatic disease and elevated overall mortality. Urethral involvement was also related to raised overall mortality. Moreover, HPV-related tumors on the basis of histologic peculiarities related to lower rates of metastatic disease. In conclusion, HPV is a common reason for penile SCC and could be diagnosed through tumor histology and authenticated through over expression of p16 on IHC. Moreover, poor prognostic indicators are the presence of LVI, PNI, and urethral involvement, whereas HPV-related tumors on the basis of histology could have decreased risk for metastatic disease.
