Ham JC, van Meerten E, Fiets WE, et al. - Given the limited progression-free survival (PFS) benefit of methotrexate in recurrent or metastatic (R/M) squamous cell carcinoma of the head and neck (SCCHN), experts aspired to explore whether adding cetuximab to methotrexate enhances PFS. In the phase-Ib-study, patients with R/M SCCHN received methotrexate and cetuximab as first-line treatment or methotrexate alone (2:1). In the combination group, median PFS was 4.5 months vs 2.0 months in the methotrexate group. Results showed an increase in PFS in correlation with providing cetuximab with methotrexate in R/M SCCHN-patients; this improvement was observed without increased toxicity.
