LncRNA WT1-AS downregulates survivin by upregulating miR-203 in papillary thyroid carcinoma

Le F, et al. - In order to analyze the roles of lncRNA WT1-AS in papillary thyroid carcinoma (PTC), researchers undertook this investigation. They analyzed PTC and non-tumor tissues from 66 PTC patients, to determine expression levels of WT1-AS, and to compare the results by performing qPCR and paired t test, respectively. In order to gauge the influences of the overexpression of WT1-AS, miR-203 and survivin on the proliferation of IHH-4 (a human PTC cell line) cells, they performed cell proliferation (CCK-8) assay. Significant downregulation of WT1-AS in PTC, as well as its link with clinical stages, was revealed. A negative correlation of WT1-AS with survivin but its positive correlation with miR-203 was seen in PTC tissues. Reduced proliferation of PTC cells, which was induced by overexpression of WT1-AS and miR-203, and increased proliferation caused by survivin overexpression, was shown in the cell proliferation assay. Thus, cancer cell proliferation in PTC is inhibited by WT1-AS, possibly through survivin downregulation by upregulating miR-203.
