LncRNA PCBP1-AS1 correlated with the functional states of cancer cells and inhibited lung adenocarcinoma metastasis by suppressing the EMT progression

Li Z, Pan C, Wang Z, et al. - Possibly, long non-coding RNAs (lncRNAs) associated with the functional states of cancer cells have crucial roles in lung adenocarcinoma (LUAD) progression, thus, researchers herein used the CancerSEA database to screen LncRNAs that were related to the functional states of LUAD cells detected in single-cell RNA sequencing (scRNA-seq) studies. To unveil the roles of their discovered novel prognostic lncRNAs, experts used transwell and tail vein metastasis assays. Initially, 849 lncRNAs were found. Of them, 11 lncRNAs showed a significant link with the prognosis of LUAD in the TCGA database. Two of them (PCBP1-AS1 and ZSCAN16-AS1) were further validated using independent GEO datasets. In scRNA-seq analysis, a significant negative correlation of both PCBP1-AS1 and ZSCAN16-AS1 with most of the functional states of LUAD cells, particularly with metastasis, was found. Findings demonstrated that suppression of LUAD metastasis could be induced by PCBP1-AS1 via targeting the EMT (Epithelial-Mesenchymal Transition) pathway. Overall, PCBP1-AS1 might be a prognostic biomarker as well as a potential treatment target of LUAD.
