Large-scale meta-analysis of mutations identified in panels of breast/ovarian cancer-related genes — Providing evidence of cancer predisposition genes

Suszynska M, et al. - Researchers performed a comprehensive meta-analysis of 48 multi-gene panel (MGP)-based studies analyzing breast cancer (BC)/ovarian cancer (OC) patients, to estimate a reliable BC and OC risk related to pathogenic variants in the selected candidate BC/OC predisposition genes. They assessed the role of 37 genes. Overall, ~120,000 BC/OC cases and ~120,000 controls were included, in whom, the mutation frequency was compared, which guaranteed strong statistical support with high confidence for most analyzed genes. They found a significant association of 13 and 11 of the analyzed genes with an increased BC and OC risk, respectively. Compared with risk attributed to BRCA2, the risk attributed to some of these genes (eg, CDKN2A and PALB2 for BC) was similar. Substantially varied profiles of genes contributing to either BC or OC risk were observed, including genes specifically related to a high risk of OC but not BC (eg, RAD51C, and RAD51D). The knowledge-based interpretation of identified mutations was supported.
