Interleukin‐7/interferon axis drives T‐cell and salivary gland epithelial cell interactions in Sjögren’s syndrome

Rivière E, Pascaud J, Virone A, et al. - This study was sought to assess the interactions between salivary gland epithelial cells (SGECs) and T cells in primary Sjögren’s syndrome (pSS) and the role of the IL‐7/interferon (IFN) axis. From pSS and controls, primary cultured SGECs were stimulated with poly(I:C), IFNα or IFNγ. Via flow cytometry, CD25 expression was evaluated. For 4 days, salivary gland explants were cultured with anti‐IL‐7R antagonist antibody (OSE‐127) and transcriptomic analysis was conducted by applying nanostring. These data imply an IL‐7‐IFNγ amplification loop involving SGECs and T cells in pSS. The data revealed that IL‐7 was secreted by SGECs stimulated with type 1 or type 2 IFN and, in turn, activate T cells that secrete type 2 IFN. In T cells, an anti‐IL‐7R antibody reduced the IFN signature during pSS and could be of therapeutic interest.
