Inhibition of fascin in cancer and stromal cells blocks ovarian cancer metastasis

McGuire S, et al. - Researchers focused on fascin, its expression pattern and function, in cancer and stromal cells of ovarian cancer (OvCa) tumors. In human cells and tissues, assessment of fascin expression was done using immunohistochemistry and immunofluorescence. They evaluated the function of fascin with in vitro functional assays, an ex vivo colonization assay and in vivo metastasis assays using siRNA/shRNA and an inhibitor. They found higher fascin expression in the stromal cell vs cancer cell compartment of ovarian tumors. The favorable prognosis for non-serous OvCa patients was suggested by low expression of fascin in the cancer cells of the primary tumor. Reduced migration of cancer and stromal cells was evident in vitro because of both knockdown and pharmacologic inhibition of fascin. Pharmacological inhibition of fascin in both cancer and stromal cells of the OvCa tumor microenvironment may potentially have a therapeutic impact.
