Identification of novel loci and new risk variant in known loci for colorectal cancer risk in East Asians

Lu Y, Kweon SS, Cai Q, et al. - Since only a small proportion of the familial risk of colorectal cancer is explained by risk variants known so far for this cancer, and this is especially noted in Asians, so, researchers analyzed 23,572 colorectal cancer cases and 48,700 controls in a genome-wide association study (GWAS) of colorectal cancer among East Asians. New risk loci were explored by selecting 60 promising risk variants for replication utilizing data from 58,131 colorectal cancer cases and 67,347 controls of European descent. Experts carried out conditional analyses in East Asians to define additional risk variants in established colorectal cancer loci. They identified the link of an indel variant, rs67052019 at 1p13.3, with colorectal cancer risk at P = 3.9 × 10–8 in Asians. Three new risk loci and two highly suggestive loci for colorectal cancer risk were discovered in this large GWAS. Evidence was generated for likely roles of multiple genes and pathways in the causation of colorectal cancer. The existence of additional risk variants in many colorectal cancer risk loci recognized earlier was also shown.
