Identification of novel loci and new risk variant in known loci for colorectal cancer risk in East Asians

Lu Y, Kweon SS, Cai Q, et al. - A genome-wide association study (GWAS) of colorectal cancer (CRC) in East Asians involving 23,572 CRC cases and 48,700 controls was conducted in order to recognize novel risk loci and new risk variant in known loci for CRC risk in East Asians. In Asians, an indel variant, rs67052019 at 1p13.3, was discovered to be related to CRC. Of the remaining 59 variants, 12 exhibited a relationship in the European-ancestry study, including rs11108175 and rs9634162 and two variants with a correlation near the genome-wide significance level (rs60911071, rs62558833) in the combined analyses of Asian- and European-ancestry data. Moreover, using data from East Asians, 13 new risk variants at 11 loci reported from former GWAS were determined. In conclusion, in this large GWAS study, three novel risk loci and two highly suggestive loci for CRC risk were recognized and evidence for possible roles of multiple genes and pathways in the etiology of CRC was presented. Thus, to enhance the understanding of the genetic basis for CRC risk, the study gives novel data.
