High calpain-1 expression predicts a poor clinical outcome and contributes to tumor progression in pancreatic cancer patients

Yu LM, et al. - The association between calpain-1 and pancreatic cancer (PC) was examined, using 96 pancreatic cancer samples and paired adjacent non-cancerous specimens. Using immunohistochemistry, calpain-1 protein expression was evaluated. In six PC cell lines, calpain-1 protein levels were evaluated by western blot (WB). PC tissues and cells exhibited overexpression of calpain-1. Association of calpain-1 overexpression, with tumor site, metastasis, and TNM stage, but not with histological grade, age, sex, and lesion size, was evident. An increased overall survival (OS) was observed in the low calpain-1 expression group vs patients expressing high calpain-1 levels. Besides, successful silencing of calpain-1 in PC cells by liposome-mediated RNA interference was noted, which led to attenuated cell growth, invasion, and metastasis in PC cells, with no effect on apoptosis. Based on the findings, calpain-1 was recommended to be considered a potential biomarker for PC prognosis and therapy.
