Family history of prostate cancer and the incidence of ERG- and PTEN-defined prostate cancer

Hashim D, et al. - Among men (n=44,126) from the prospective Health Professionals Follow-up Study, researchers assessed the links between family history and the incidence of molecular subtypes of prostate cancer (PCa), including two somatic genetic aberrations: fusions of androgen-regulated promoters with ERG and, separately, PTEN loss. They performed immunohistochemistry to evaluate ERG and PTEN status. For links between self-reported family history of PCa and molecular subtypes of disease, they estimated hazard ratios and 95% confidence intervals using multivariable competing risks models. Findings are suggestive of a possible positive association of PCa family history with PCa in all ERG and PTEN subtypes, this indicates the role of genetic susceptibility in their development. Particularly, ERG-negative disease could be related to positive family history.
