Emergence of the Asian lineage of Zika virus in Angola: An outbreak investigation

Hill SC, Vasconcelos J, Neto Z, et al. - The emergence and circulation of Zika virus in Angola were investigated. Employing real-time reverse transcription PCR (RT-PCR), the Instituto Nacional de Investigação em Saúde (Ministry of Health, Luanda, Angola) tested diagnostic samples collected by the Angolan Ministry of Health as part of routine arboviral surveillance. In addition, researchers tested samples from a 2017 study of people with HIV in Luanda for further samples positive for Zika virus and appropriate for genomic sequencing. In this multi-component outbreak investigation, the first known outbreak of Asian lineage Zika virus in continental Africa, they tested serum samples from 54 people with suspected acute Zika virus infection, 76 infants with suspected microcephaly, 24 mothers of infants with suspected microcephaly, 336 patients with suspected dengue virus or chikungunya virus infection, and 349 samples from the HIV study. They obtained data for five cases of Zika virus infection between December, 2016, and October, 2017, which were confirmed by real-time RT-PCR, Although individually, the surveillance, genetic, and clinical data were limited, together these independent lines of evidence produce a cohesive picture of the outbreak of Zika virus infection in Angola. The three Zika virus genomes, that were sequenced, describe the first Asian lineage genomes inscribed from Africa, and the first genome isolated from a case with Zika-virus-associated microcephaly in Africa. As per phylogenetic analyses, Zika virus was apparently introduced to Angola from Brazil and then circulated for 17–28 months. Finally, the serological investigations in one mother-child pair backed the view that the Zika virus outbreak produced microcephaly in Angola.
