EGFR mutations compromise hypoxia-associated radiation resistance through impaired replication fork-associated DNA damage repair

Saki M, et al. – This study aimed at determining whether epidermal growth factor receptor (EGFR) mutations compromised hypoxia–associated radiation resistance through impaired replication fork–associated DNA damage repair. Within an altered DNA damage response of hypoxic non-small cell lung carcinomas (NSCLC) cells, mutant EGFR expression, or EGFR blockade by cetuximab exerted a synthetic lethality effect. Also, it significantly compromised radiation resistance in hypoxic tumor cells.
