Effect of clozapine dose and concentration on fasting concentration of appetite regulating peptides

Margulska A, et al. - In this study, the researchers investigated the effect of clozapine dose and concentration on fasting concentration of appetite regulating peptides: Cocaine- and amphetamine-regulated transcript (CART), peptide YY (PYY(1–36)), neuropeptide Y (NPY), agouti-related protein (AgRP), des-acylated ghrelin, leptin, and obestatin. No direct association was observed between serum concentration of clozapine with increased or decreased level of appetite-regulating peptides.
Methods
- The researchers analyzed 30 subjects with schizophrenia on clozapine monotherapy.
- They measured serum concentration of clozapine and fasting serum levels of des-acylated ghrelin, NPY, AgRP, PYY(1–36), CART, leptin and obestatin.
Results
- While NPY concentration was negatively associated with clozapine concentration (r = -0.55, p=0.01), leptin concentration was negatively correlated with clozapine dose (r = -0.53, p=0.002).
- As per findings, correlations with other peptides were not significant.
