Double-edged sword of inflammasome genetics in colorectal cancer prognosis

Cambui RAG, do Espírito Santo GF, Fernandes FP, et al. - Given inflammation represents a hallmark of colorectal cancer (CRC) and there can be a beneficial or detrimental role of inflammasome-dependent cytokines IL-1ß and IL-18 in tumorigenesis based on cancer type, and variants in inflammasome genes were related to tumor development and/or outcome, and have been suggested as likely biomarkers for population screening, so, in order to gain insights, researchers undertook this study with 215 CRC patients, with a follow-up for 10 years. The participants were investigated for 9 polymorphisms in selected inflammasome genes. To assess the link between the polymorphisms and CRC prognosis, experts conducted multivariate association analysis and survival analysis. Findings revealed that CRC prognosis was detrimentally influenced by variants related to lower levels of IL-18 (rs1834481, rs5744256) or those associated with increased activation of inflammasome receptors NLRP1 (rs12150220) and NLRP3 (rs35829419), and these variants may serve as prognostic markers.
