Diffuse high-grade gliomas with H3 K27M mutations carry a dismal prognosis

Karremann M et al. – In the current study, the diffuse midline glioma, H3 K27M-mutant, was shown to be a distinct entity corresponding to WHO grade IV, which has a universally fatal prognosis.
Methods
- 85 diffuse gliomas were characterized based on tumor site, histone 3 mutational status, WHO grade, age, gender, and extent of tumor resection.
Results
- K27M-mutant gliomas had an aggressive clinical course independent of anatomic location.
- The H3 K27M mutation was the only independent parameter predictive of overall survival.
- Among H3 K27M-mutant tumors, anatomic midline location, histopathological grading, and extent of tumor resection had no influence on survival.
