Determination of the 90% effective dose of intranasal dexmedetomidine for sedation during electroencephalography in children

Liu H, et al. - Researchers used a biased coin design up-and-down sequential method (BCD-UDM) in this double-blind trial to determine, in children under 3 years old, the 90% effective dose (ED90) of intranasal dexmedetomidine (DEX). Participants were 53 children under 3 years old who were undergoing EEG, for which DEX was required. They administered 2.5 μg kg-1DEX to the first patient and, based on the response of the previous patient, the dose of DEX for following patient was decided. They used isotonic regression to estimate the ED90 after analyzing patient responses. They defined 3.28 µg kg-1 as the 90% effective dose of intranasal DEX sedation for EEG in children under 3 years old. The estimated 95% CI was 2.74 ~ 3.39 µg kg-1. No significant adverse events were observed.
