Comprehensive analysis of IgA nephropathy expression profiles: Identification of potential biomarkers and therapeutic agents

Gholaminejad A, Gheisari Y, Jalali S, et al. - Researchers applied an integrative approach to mRNA and miRNA expression profiles in peripheral blood mononuclear cells in order to find out a gene signature as well as novel potential targets/biomarkers in IgA nephropathy (IgAN). From gene expression omnibus database, datasets were selected. They analyzed “GSE25590” (miRNA) and “GSE73953” (mRNA) datasets, and discovered 628 intersecting mRNAs post-integration. Mainly a link with “Innate immune system”, “Apoptosis”, as well as “NGF signaling” pathways was shown by mRNAs. Experts built a multilayer regulatory network and introduced various hub-differentially expressed genes [Tp53, STAT3 (signal transducer and activator of transcription 3), Jun, etc.], differentially expressed miRNAs (miR-124, let-7b, etc.), transcription factors (NF-kB, etc.), and long-non coding RNAs (HOTAIR, etc.) as potential factors in IgAN pathogenesis. Overall, a deeper insight into IgAN pathogenesis as well as introduction of various key molecules as potential treatment target/non-invasive biomarkers was offered by combination of two different expression datasets as well as creation of a multilayer regulatory network in this study.
