Comparison of the efficacy and safety of minocycline and clofazimine in chronic and recurrent erythema nodosum leprosum – A randomized clinical trial

Hanumanthu V, Thakur V, Narang T, et al. - According to this prospective randomized clinical trial, minocycline demonstrated superior efficacy to clofazimine in terms of rapid and sustained improvement of erythema nodosum leprosum (ENL) episodes with fewer adverse events.
The sample consisted of 60 participants with chronic/recurrent ENL.
Participants in the study were randomly assigned (1:1) to receive either minocycline 100 mg once daily or clofazimine 100 mg thrice daily for 12 weeks along with prednisolone according to WHO protocol, and they were followed up for 6 months.
When compared with the clofazimine group, the minocycline group achieved initial control of ENL sooner.
During the study period, the number of participants experiencing ENL flares/recurrences was comparable in both groups.
When compared with the clofazimine group, participants in the minocycline group remained in remission for a longer period of time after the initial control of ENL.
In both groups, the mean additional prednisolone dose required to control ENL flares/recurrences was comparable.
There were fewer side effects in the minocycline group than in the clofazimine group.
