Comparison of normal and metaplastic epithelium in patients with stable vs persistently symptomatic severe asthma using laser-capture microdissection and data-independent acquisition–mass spectrometry

Weitoft M, Müller C, Åhrman E, et al. - Unbiased label-free quantitative proteome analyses were conducted on laser-capture–microdissected areas of specific epithelial structures from people with severe asthma (SA) with differing degrees of symptom control in order to investigate the use of laser-capture microdissection of specific epithelial structures combined with quantitative data-independent acquisition mass spectrometry to illustrate variations in protein composition in individuals with SA with varying symptom control. A total of 1,993 and 1,652 and 1,458 and 1,647 stable SA and symptomatic SA proteins, in normal epithelium and in the metaplastic epithelium, respectively, were quantified. New treatment approaches are required for people with severe asthma and exploratory studies of unbiased nature like this may aid while looking for new mechanisms and potential targets involved in the disease pathology.
