Cognitive function in metformin exposed children, born to mothers with PCOS – follow-up of an RCT

Greger HK, et al. - This is the first follow-up of cognitive function in a placebo-controlled randomized controlled trial of children exposed to metformin in utero. In this follow-up of two randomized, placebo-controlled studies, researchers ascertained if metformin exposure in utero had any impact on offspring cognitive function. These investigations were carried out at 11 public hospitals in Norway. From the first trimester to delivery, participants were randomized to metformin 1,700 and 2,000 mg/d or placebo. Parents of 292 children were invited to give permission for their children to partake. Between April 2014 to June 2016, a total of 93 children (52 in the metformin group and 41 in the placebo group) were tested. There was no evidence of long-term impact of metformin on average child cognitive function. Increasing borderline intellectual functioning in children exposed to metformin needs to be viewed with caution due to the small sample size.
