Clinical factors associated with urinary tract cancer in individuals with Lynch syndrome

Wischhusen JW, Ukaegbu C, Dhingra TG, et al. - Via a cohort of 52,758 consecutively determined individuals who underwent Lynch syndrome (LS) testing at a commercial laboratory, experts recognized clinical factors related to urinary tract cancers (UTC) among LS carriers. LS carriers, in comparison with non-carriers, were significantly more inclined to have had UTC. LS-associated UTC was independently related to the male gender, progressed age, the familial burden of UTC, and pathogenic EPCAM/MSH2 variants however not MLH1 variants, race, or history of other LS-associated malignancy. A total of 143/158 LS carriers with UTC had ≥ 1 of the following features— male gender, EPCAM/MSH2 variants, or family history of UTC and a total of 1,236/1,251 LS carriers lacking all of these features had no history of UTC. Hence, certain clinical factors could assuredly recognize LS carriers most expected to be at risk for UTC. Moreover, a predictable subset of LS carriers may be most suitable to profit from UTC surveillance/prevention.
