Characterization and function of tumor necrosis factor alpha and interleukin‐6‐induced osteoclasts in rheumatoid arthritis

Yokota K, Sato K, Miyazaki T, et al. - This study was sought to verify the characterization and function of human TNFα and IL‐6‐induced osteoclasts using peripheral blood collected from patients with rheumatoid arthritis (RA) and healthy donors. Peripheral blood monocytes were cultured with TNFα and researchers assessed IL‐6 or RANKL, and their bone resorption ability. They analyzed the impacts of NF‐AT inhibitor and JAK inhibitor. Moreover, in patients with RA, the correlation between the number of TNFα and IL‐6‐induced osteoclasts or osteoclasts differentiated from peripheral blood mononuclear cells (PBMCs) and the modified total Sharp score (mTSS) or the whole‐body bone mineral density (BMD) were explored. This study’s findings revealed that TNFα and IL‐6‐induced osteoclasts may contribute to the pathology of inflammatory arthritis correlated with joint destruction, such as RA.
