Association of TRAP1 with infliximab-induced mucosal healing in Crohn's disease
Journal of Gastroenterology and Hepatology Jun 22, 2019
Park SH, et al. - In patients with Crohn's disease (CD), researchers examined the genetic factors correlated with response to anti-tumour necrosis factor (TNF) therapy, such as infliximab (IFX). A two-stage genome-wide association study (GWAS) was carried out to identify loci influencing the response to IFX among Korean CD subjects, consisting of 42 good mucosal healing responders and 70 non-responders. TRAP1 (TNF receptor associated protein 1) functional significance was investigated utilizing dextran sodium sulfate-induced colitis model in TRAP1 transgenic mice. TRAP1 transgenic mice exhibited a better response to IFX compared to the wild-type mice in the dextran sodium sulfate-induced acute colitis. In CD patients following IFX therapy, the TRAP1 gene was linked to mucosal healing. Identifying mucosal healing genetic predictors to anti-TNF therapy may prevent subjects from being exposed to ineffective therapies.
