Analysis of threshold change of tumor mutation burden in gastric cancer

Journal of Oncology Jul 26, 2021

He X, Yu M, Wang X, et al. - Researchers examined how tumor mutation burden (TMB) in gastric cancer (GC) has changed and determined its correlation with prognosis. In all 262 GC patients included in this study, advanced stage was detected and treatment included surgical removal of D2 lymph nodes and dissection. The total mutation rate was 92.85% among these patients. Following were the top 5 mutant genes: TP53, RB1, ARID1A, KMT2B, and RET. Statistically significant expression level of TMB was noted in GC patients with age, drinking history, and differentiation type. Univariate and multivariate logistic analysis indicated statistically significant correlation of age, drinking history, clinical stage, differentiation type, and TMB with the overall survival rate of GC patients. Overall findings suggest that changes in TMB frequently accompany GC patients, and its expression level is noted to be closely related to the degree of pathological differentiation, which is an independent factor influencing the prognosis of GC patients. They suggest a possible utility of high TMB value for evaluating the prognosis and providing a reference for the formulation of clinical treatment plans for GC patients.
