A polygenic risk score to predict future adult short stature amongst children

Lu T, Forgetta V, Wu H, et al. - In the present study, the researchers sought to develop a polygenic risk score for adult height and assess its clinical utility. Based on a meta-analysis of genome-wide association studies, a polygenic risk score was developed and tested on the Avon Longitudinal Study of Parents and Children (ALSPAC) cohort. Candidates involved 442,599 genotyped White British individuals in the UK Biobank, and 941 genotyped child-parent trios of European ancestry in the ALSPAC cohort. In European ancestry populations, a polygenic risk score may be used instead of or in addition to mid-parental height to boost screening for children at risk of developing short stature in adulthood.
