2-methoxyestradiol and its combination with a natural compound, ferulic acid, induces melanoma cell death via downregulation of Hsp60 and Hsp90

Kamm A, Przychodzeń P, Kuban–Jankowska A, et al. - Using the A375 melanoma cellular model, researchers tested 2-methoxyestradiol (potent chemotherapeutic) with respect to its anticancer potential. They merged the treatment with a naturally occurring polyphenol, ferulic acid, to further explore the anticancer efficacy of 2-methoxyestradiol. They used the melanoma A375 cellular model to get the results. A partial association of the molecular mechanism of action of 2-methoxyestradiol and ferulic acid, with the attenuation of Hsp60 and Hsp90 (heat shock proteins) levels and the induction of nitric oxide in the A375 melanoma cell model, was evident, while either separate or combined treatment of 2-methoxyestradiol and ferulic acid did not cause any alteration in Hsp70 expression. In case of chemoresistance mechanisms, its significance is particularly emphasized as Hsp70 accumulation attenuated induction of cancer cell death, therefore, reducing antitumour efficacy.
